سنتز، شناسایی، مطالعات برهمکنش CT-DNA و محاسبه پارامترهای ترمودینامیکی پلاتین شامل اتم های دهنده N2S2

چکیده
پلاتین (IV )شامل لیگاند 2-مرکاپتوبنزوتیازول دی سولفید (MBTS) با یک پیوند دی سولفیدی از طریق واکنش) PtAr2(SMe2)2] (Ar=Ph] با 2-مرکاپتوبنزوتیازول دی سولفید انجام شده است. همچنین کمپلکس شامل گروه های Ar = p-tolyl سنتز شده و خواص بیوشیمی ان ها با یکدیگر مقایسه شد. در این تحقیق 2–مرکاپتوبنزوتیازول (MBT) در طی شکستن پیوند از MBTS تشکیل شده که به عنوان لیگاند کی لیت شامل گوگرد- نیتروژن عمل می کند. ترکیبات سنتز شده با استفاده از روش های اسپکتروسکوپی مختلف مانند FT-IR, 1HNMR آنالیز عنصری و طیف های UV/Vis شناسایی شدند. علاوه بر این بر همکنش ترکیبات سنتز شده و DNA با استفاده از تکنیک های مختلف مانند طیف سنجی های یووی و فلورسانس و بررسی های ویسکومتری انجام شد. ثابت های پیوندی Kb و Kfبرای کمپلکس های پلاتین در دماهای مختلف محاسبه و داده های بدست آمده با یکدیگر مقایسه شدند. پارامترهای ترمودینامیکی بر همکنش های کمپلکس های پلاتین و DNA محاسبه شدند. مقادیر Kb نشان می دهند که کمپلکس های پلاتین سنتز شده به مقدار قابل توجهی با DNA برهمکنش می دهند. مقادیر مثبت انتالپی و انتروپی نشان می دهد که برهمکنش ها از نوع هیدروفوبیک می باشد. ویسکوزیته DNA با افزایش غلظت کمپلکس ها افزایش می یابد که نشان دهنده برهمکنش DNA با این ترکیبات است. داده های بدست آمده نشان دادند که مکانیسم برهمکنش ها از نوع ایترکیلیشن می باشد. مقایسه ثابت برهمکنش ها نشان می دهند که بر همکنش پلاتین شامل گروههای دی فنیل بیش تر از پلاتینی است که دارای گروههای پارا تولیل است

چکیده English

Platinum(IV) containing 2-mercaptobenzothiazole disulfide (MBTS) ligand with a disulfide bond was synthesized through the reaction of [PtAr2(SMe2)2] (Ar = Ph) with 2-mercaptobenzothiazole disulfide. Also, the complex containing Ar = p-tolyl groups was synthesized and their biochemical properties were compared. In this research, 2-mercaptobenzothiazole (MBT) was constructed during the breaking of the S–S bond in 2-mercaptobenzothiazole disulfide (MBTS), which then act as a sulfur-nitrogen chelating ligand. The synthesized organometallic compounds were identified using various spectroscopy methods such as FT-IR, 1HNMR, elemental analysis, and electronic spectra. Furthermore, the interactions of synthesized compounds and DNA (deoxyribonucleic acid) were studied using different techniques such as electronic and photoluminescence spectroscopy and viscosity investigations. The intrinsic binding constants containing Kb and Kf were calculated for Pt complexes at different temperatures, and the obtained data were compared with each other. The spectra of emission and absorption changed upon the addition of DNA. Moreover, thermodynamic parameters of the interaction between Pt compounds and deoxyribonucleic acid were obtained. The resulting Kb values confirmed the Pt complexes considerably interacted with DNA. The obtained positive changes in enthalpy and entropy confirmed the corresponding hydrophobic interaction. The viscosity of deoxyribonucleic acid was increased during the addition of complexes, indicating the interaction with these compounds. The obtained data indicated the mechanism of intercalative for CT-DNA interactions with Pt complexes. Moreover, comparing the intrinsic binding constants data showed that the interaction of the organoplatinum complex containing phenyl groups with DNA is more than its analogue containing para-tolyl groups.
